  • Phase 3 TRIUMPH-1 (Lilly, May 2026): 2,339 adults; mean weight reduction of 28.3% at 80 weeks on 12 mg, versus 2.2% on placebo.
  • Phase 3 TRIUMPH-2 and TRIUMPH-3 (Lilly, July 2026): 80 weeks on 12 mg; 20.8% vs 4.0% on placebo in adults with type 2 diabetes (n=1,152) and 22.6% vs 3.2% in adults with severe obesity and cardiovascular disease (n=1,949).
  • Lilly says it plans to submit retatrutide to the FDA in the first quarter of 2027.

Coskun et al., Cell Metabolism described LY3437943 as a 39-residue agonist at GIP, GLP-1 and glucagon receptors, with preclinical and early clinical evidence. This establishes research context for the molecule, not a test result for the product batch.

2023: randomized Phase 2 obesity trial

Jastreboff et al., NEJM studied 338 adults. At 48 weeks, mean weight change in the 12 mg study arm was −24.2%, versus −2.1% with placebo. The primary endpoint was at 24 weeks. Gastrointestinal effects were common, and heart-rate increases were reported. Results cannot be assumed for another population or product.

2024: liver-fat substudy

Sanyal et al., Nature Medicine assessed 98 participants with elevated liver fat. MRI measurements improved in the study groups. This was a liver-fat endpoint, not evidence that retatrutide cures cirrhosis or prevents liver-related death.

June 2026: published Phase 3 TRANSCEND-T2D-1

Bajaj et al., The Lancet reported a placebo-controlled diabetes trial with HbA1c as its primary endpoint. The research guide explains its results and analysis method. Assess the trial medicine and a commercial research vial separately; publication does not verify a retail batch.

June and July 2026: TRIUMPH sponsor reports

Lilly’s June TRIUMPH-1 update reported 28.3% mean weight reduction in the 12 mg arm at 80 weeks. Its 104-week extension involved a selected population and should be identified separately. July TRIUMPH-2 and TRIUMPH-3 results concerned different populations with diabetes or severe obesity. These are sponsor-reported results, not a direct comparison with tirzepatide.

August 2026: cardiovascular risk biomarkers

The Diabetes, Obesity and Metabolism analysis examined lipid and inflammatory markers from two earlier Phase 2 trials. It was a post-hoc analysis. Changes in risk markers do not establish fewer heart attacks, strokes or cardiovascular deaths.
